Benfica

Semedo came up through Sintrense's youth system, making his first-team debut at the age of 17. He and Manuel Liz signed five-year contracts with Benfica on January 12, 2012, which began on July 1, 2012. Both played a season on loan in the third tier with Fátima.

In the 2013 off-season, Semedo returned to Benfica, where he was assigned to the B-team in the Segunda Liga and made his professional debut on August 10 in a 0–0 away draw against Trofense. He was tipped as a prospective replacement for long-time first-team incumbent Maxi Pereira after nearly 60 appearances for the reserves. Semedo was a UEFA Champions League player for Benfica.

Semedo's contract was extended until 2021 after Pereira's departure, and he joined the first team on their pre-season tour of North America in July 2015. He made his first appearance for the first team on August 9, 2015, in a 0–1 loss to

Sporting CP

in the Supertaça Cândido de Oliveira. A week later, he scored his first goal for them in a 4–0 home triumph over Estoril in the Primeira Liga.

After establishing himself in the starting lineup, Semedo suffered an injury while playing for the Portugal national team in October 2015. His right knee injury necessitated surgery, and he was projected to be out for two months. Barcelona signed an agreement with Benfica for Semedo's joining on July 13, 2017, pending medical checks. He signed a five-year contract the next day, with the Portuguese club earning €30.5 million and a further €5 million if he participated in 50 matches for his new squad. On August 16, he made his debut against Real Madrid in the second leg of the Supercopa de Espana, losing 0–2 at the Santiago Bernabéu Stadium.

On September 13, 2017, Semedo made his Champions League debut for the club, and he was praised for his performance in the 3–0 group stage triumphs over

Semedo signed a three-year contract with English Premier League club Wolverhampton Wanderers on September 23, 2020, for an initial transfer price of €30 million (about £27.5 million, with a potential further £9.2 million in add-ons) paid by Wolves.

Semedo made his Wolves debut and first Premier League match in a 4–0 loss to West Ham United on September 27, 2020. On 4 October 2020, he made his Molineux debut for Wolves in the team's next Premier League encounter, a 1–0 win over Fulham.

He scored his first Wolves goal in Nuno Espirito Santo's farewell game in charge, a 1–2 home defeat to
