Piola started his career with Pro Vercelli, the Italian club team, making his Serie A debut against Bologna on 16 February 1930, scoring 13 goals at the age of 17 in his first year. Piola scored six goals on 29 October 1933, the joint-most goals scored in a single Serie A match, in a 7-2 victory over Fiorentina. In 127 games for Pro Vercelli in Serie A, he went on to score 51 goals.

Considered one of the best strikers of all time, during his career, Piola was well recognized for his goalscoring abilities and his talent for the target.

During his time, he was known as a modern and well-rounded player, as he used his physical features, intellect, and control to play with his back to goal and lay off the ball for teammates, and provide them with assistance.
